ABSTRACT

Objective:To investigate the effects of human placenta tablets combined with active folic acid and compound nutrient tablets on Th1 cytokines and hormone levels in pregnant patients with threatened miscarriage. Methods:A total of 226 pregnant patients with threatened miscarriage who received treatment in Linyi Traditional Chinese Medicine Hospital from April to September 2021 were included in this study. They were randomly divided into a control group and an observation group, with 113 patients in each group. The control group was treated with human placenta tablets. The observation group was treated with human placenta tablets combined with active folic acid and compound nutrient tablets. All patients were treated for 4 weeks. Before and after treatment, serum levels of Th1 cytokines, β-human chorionic gonadotropin, progesterone, and estradiol as well as the rate of success of protection against miscarriage, adverse reactions, and perinatal complications were compared between the two groups. Results:After 4 weeks of treatment, serum levels of interleukin-2, tumor necrosis factor-α, and interferon-γ in the observation group were significantly lower than those in the control group ( t = 27.53, 20.99, 31.69, all P < 0.001). Serum levels of β-human chorionic gonadotropin, progesterone, and estradiol in the observation group were (143.79 ± 9.56) IU/L, (36.43 ± 4.71) ng/L, (234.72 ± 13.29) pmol/L, which were significantly higher than (122.53 ± 7.47) IU/L, (29.32 ± 4.22) ) ng/L, (167.86 ± 8.93) pmol/L in the control group ( t = 18.63, 11.95, 44.39, all P < 0.001). The rate of success of protection against miscarriage in the observation group was significantly higher than that in the control group (92.9% vs. 76.1%, χ2 = 12.20, P < 0.05). There were no significant differences in the incidences of adverse reactions and perinatal compilations between the two groups (both P > 0.05). Conclusion:Human placenta tablets combined with active folic acid and compound nutrient tablets can improve serum levels of Th1 cytokines and hormones, and increase the rate of success of protection against miscarriage, without increasing the incidences of adverse reactions and perinatal complications.

ABSTRACT

Preterm birth and its complications are the leading cause of neonatal death, and also a perinatal problem that contributes to the global burden of disease. Recently, the association between maternal heat exposure and elevated risk of preterm birth has been found in lots of studies. But the potential mechanisms of how heat exposure increase the incidence of preterm birth are still unclear. Thus, we reviewed maternal vulnerability factors and territorial moderators associated with preterm birth due to heat exposure during pregnancy, and summarized potential mechanisms between heat exposure and risk of preterm birth based on previous studies. We found that heat exposure during pregnancy may involve various mechanisms to increase the risk of preterm birth, such as oxidative stress, inflammation, reproductive and urinary infections, neuroendocrine changes, and complications during pregnancy. Prospective cohort studies and animal experiments should be conducted to clarify the adverse health effects of heat exposure on different types of preterm birth from three facets of "exposure-mechanism-effect", aiming to provide a scientific basis for the protection of maternal and infant health through conducing clinical preventive interventions against preterm birth in scorching weather.

ABSTRACT

Objective:To explore the impact of social support on cognitive function and depression in patients with mild cognitive impairment (MCI).Methods:From March to September 2018, 5 765 subjects over 60 years old from 52 villages in Yanlou Town, Yanggu County were selected and they were screened by mini-mental state examination (MMSE) and activities of daily living(ADL). Finally 4 750 valid questionnaires were recovered.According to the " Diagnostic and Statistical Manual of Mental Disorders" 4th Edition (DSM-Ⅳ), 733 patients with MCI (patient group) and 3 662 patients with normal cognitive function (healthy control group) were diagnosed. The social support rating scale (SSRS) and geriatric depression scale-15 (GDS-15) were used to evaluate the patients.SPSS 26.0 software was used for independent sample t-test, chi-square test, Pearson correlation analysis and linear regression analysis. Results:The total score of social support (48.55±9.72), objective social support (16.49±4.00), subjective social support (24.28±4.75) and social support utilization (7.78±2.85) in patients group were significantly lower than those in the healthy control group (total score of social support (50.94±7.66), objective social support (17.23±3.42), subjective social support (25.59±3.61) and social support utilization (8.13±2.71)). The differences were statistically significant ( t=-6.291, -4.363, -8.245, -3.068, all P<0.05) .All the dimensions of social support(total score, objective support, subjective support, support utilization) were positively correlated with cognitive function ( r=0.084, 0.062, 0.128, 0.011, all P<0.05), and negatively correlated with depression score ( r=-0.240, -0.195, -0.200, -0.169, all P<0.01). Subjective social support, objective social support and social support utilization could positively predict MMSE score of MCI patients( β=0.190, 0.007, 0.029, all P<0.05), while could negatively predict the GDS-15 score of MCI patients( β=-0.145, -0.098, -0.105, all P<0.05). Conclusion:Good social support is a protective factor for cognitive function and depression in MCI patients.

ABSTRACT

Objective To study the effect of cyclophosphamide(CP) and its metabolites acrolein(ACR) on PTEN gene deleted on chromosome 10 after acting on ovarian cancer cellsSKOV3.Methods Different concentrations of CP and ACR were selected to act on recombinant PTEN protein. The phosphorylation activity of PTEN was detected by PNPP.The expression of PTEN protein was detected by Western blot. The binding mode of drug with protein was detected by the biotin combined with protein;meanwhile the expression change of P53/TP53 in PTEN gene pathway was analyzed. The target protein was obtained by immunoprecipitation(IP) after different drug concentrations acting on the cells. The phosphorylation activity of the target protein was detected by high performance liquid chromatography(HPLC).Results After the drug metabolites acting on recombinant PTEN protein, the phosphorylation activity was decreased with the increase of drug concentration, while the expression of ACR antibody action was increased with the drug concentration elevation. The expression of protein and biotin in different experimental groups was increased with the increase of drug concentration. The PTEN phosphorylation activity was decreased with the drug concentration increased in cells, and so did the expression of TP53 protein. Conclusion CP metabolite ACR induces the cytotoxicity by inhibiting PTEN protein phosphorylation activity.

ABSTRACT

Inflammatory bowel disease(IBD) is characterized by chronic non-specific inflammatory dis ease. The inflammation caused by abnormal immune response in intestinal mucosa exerts an important role in the pathogenesis of IBD.T helper 17 cells(Th17)are involved in the development of chronic inflammation and auto immune disease, whereas regulatory T cells can inhibit the function of autoimmune, and thus the balance of Th17/Treg balance plays an important role in intestinal homeostasis.Various strategies can be done to modulate Th17/Treg balance, such as antigen-presenting cells, microoganisms, some key molecules, natural compounds, proving a promising therapeutic method for IBD.

ABSTRACT

OBJECTIVE To evaluate the anti-vertigo effect of phencynonate hydrochloride. METHODS To detect the improvement of phencynonate hydrochloride on cerebral blood flow, a rat model wa s es?tablished with bilateral common carotid arteries occlusion. Phencynonate hydrochloride 0.1-4.0 mg·kg-1 was ig given, twice a day for three consecutive days and the alteration of cerebral blood flow was measured with laser Doppler flowmetry. Rotating acceleration equipment was used to provocate mouse vertigo for 30 min, and the spontaneous locomotor activities were tested for occurrence of vertigo in mice. Phencynonate hydrochloride 1.4-5.6 mg·kg-1 was ig given before rotating acceleration. Gastric phenol red emptying rate was used to determine the anti-nausea effect of the test drug in mice 30 min after phencynonate hydrochloride 1.4-8.4 mg·kg-1 was ig given. RESULTS The cerebral blood flow of the rat model with bilateral common carotid arteries occlusion was reduced significantly after 24 h (P<0.01). Compared with the model group, phencynonate hydrochloride (0.5, 2.0 and 4.0 mg · kg-1) increased the cerebral blood flow in a dose-dependent manner in rats with cerebral ischemia (P<0.01). The spontaneous locomotor activities were significantly reduced after vertigo stimulation in mice (P<0.05). Compared with the model group, phencynonate hydrochloride (2.8 and 5.6 mg · kg-1) increased the movement distance and speed of vertigo mice (P<0.05). Phencynonate hydrochloride (2.8, 5.6 and 8.4 mg·kg-1) inhibited the gastric emptying of mice (P<0.05). CONCLUSION Phecynonate hydrochloride can improve the cerebral blood flow and locomotor activities in vertigo rats, while inhibiting gastric emptying, which points to the therapeutic potential of phencynonate hydrochloride for vertigo in clinic.

ABSTRACT

OBJECTIVE:To guide rational use of Heamocoagulase agkistrodon for injection, and to provide reference for phar-maceutical care. METHODS:Treatment methods and outcome of 9 cases of allergic shock induced by Heamocoagulase agkistrodon for injection were retrieved from CNKI and VIP database during 2009-2015,and then analyzed statistically, based on one case of allergic shock induced by Heamocoagulase agkistrodon for injection in the Affiliated Hospital of Zhunyi Medical College. RE-SULTS:10 pationts with allergic shock induced by Heamocoagulase agkistrodon for injection mainly used the drug for hemostasis before and after surgery, heavy menstrual bleeding, etc.,with dose of 1 or 2 U. Route of administration included intravenous injec-tion and intravenous dripping. Allergic shock often occurred within 30 min after medication. CONCLUSIONS:The use of Heamoco-agulase agkistrodon for injection in some patients belongs to off-label drug use,and route of administration is different from that stated in package inserts. Allergic shock induced by Heamocoagulase agkistrodon for injection occurs rapidly;the drug should be used strictly according to package inserts and authoritative literatures;emergency plan should be developed before medication in or-der to guarantee the safety of drug use.

ABSTRACT

Objective To design and create a C/S-J type of biliary self-releasing stent, and to study its safety and efficacy in preventing post-ERCP complications. Methods 118 patients with common bile duct stones treated in our hospital were enrolled into this study from October 2013 to May 2015.These patients were randomly divided into two groups:the experimental group who underwent ERCP + EST + C/S-J type of self-releasing biliary stent drainage, while the control group underwent ERCP + EST + ENBD.The incidences of post-ERCP acute pancreatitis (PEP) and cholangitis in the two groups and the time the self-releasing stent was dislodged from the biliary system in the experimental group were recorded. Results The incidence of PEP was 6.4% (5/78) and 7.5% (3/40) in the experimental and the control group, respectively (P > 0.05).There were no patients who developed postoperative acute cholangitis in the two groups. The stents were dislodged from the biliary system on the first day after the procedure in 2 patients in the experimental group without any complications. One stent failed in self-releasing but was removed successfully with endoscopy 3 months later. In the other 75 patients, the stents were successfully dislodged and were excreted outside the patient's body through the intestinal tract (mean 11.4,range 9 ~ 14) days, without any complications. Conclusion The C/S-J type of biliary self-releasing stents is safe and efficacious in preventing post-ERCP pancreatitis and cholangitis.
